Experimental procedure:
Pass the water sample through the filtration device. With a filter media packing volume of 400 ml loaded into the chromatography column (diameter 50 mm x height 300 mm, inner diameter 45 mm), the media bed height is approximately 24.5 cm. Before packing the media, place some cotton or 2-4 mm quartz sand above the outlet valve to prevent loss of fine media particles;
When packing the media, first fill the column with a certain volume of clean water or tap water, then slowly pour the media into the water. This minimizes irregular voids in the filter bed and makes the media layer compact and tight. After packing, ensure a liquid level difference of about 3 cm of clean water above the media layer, then fix the column in place.
Set the filtration rate to 5 m3/(m3.h), i.e. a peristaltic pump flow of 33.3 ml/min; calibrate the peristaltic pump flow manually with a timer. During filtration, maintain a liquid level difference of about 3-5 cm above the media as a buffer to prevent the water from directly impacting the media bed;
Start the peristaltic pump and slowly open the lower valve of the column. Replace the clean water with the target water sample; discard the first 10 min of effluent, after which it becomes the separator effluent.
Control the effluent flow through the lower valve of the column. To maintain the liquid level difference, adjust the effluent flow from time to time so that the inflow and outflow are as balanced as possible.
If a smaller chromatography column is used, packing only 100 mL of media, the column diameter is about 2.5 cm and the packing height about 20.5 cm. Set the filtration rate to 10 m3/(m3.h) (because the height-to-diameter ratio is larger, the rate can be set higher); the peristaltic pump flow should then be 16.5 ml/min.
